They will study the progress made on Asean's economic integration agenda, discuss the bloc's role in the regional political and security architecture, and receive updates on the Asean Connectivity initiative.
At the close of the summit tomorrow, they are expected to issue a statement on the sustainability of regional economic recovery and another on the climate change.
Six months ago at the last Asean Summit in Hua Hin, Thailand, the leaders said a high-level task force would be set up to draw up a plan on how best to connect the region.
According to the latest figures, Asean has a population of about 584 million and a combined GDP of US$1.5 trillion.
A draft of a statement that Asean leaders will issue tomorrow says road, sea and air links in the region should be completed more quickly to complement ongoing efforts to achieve greater integration.
Efforts to connect the region's power grid and natural gas pipelines, and to improve Internet and telecommunications connections, should also be hastened, the statement says.
Accompanying PM Lee in Hanoi are Foreign Minister George Yeo, Trade and Industry Minister Lim Hng Kiang, and Community Development, Youth and Sports Minister Vivian Balakrishnan.
While Mr Lee is away, Deputy Prime Minister and Defence Minister Teo Chee Hean will be Acting Prime Minister.
